If you have a week you can take a 7 night cruise starting in Seattle or Vancouver up to Seward/Anchorage, or go round trip from Seattle or Vancouver and do the inside passage. 
If you have a longer vacation planned you can combine a cruise and land vacation or fly directly to Alaska and take an in depth independent or escorted tour.
As far as fishing goes:  If you think Minnesota’s 10,000 lakes are great Alaska has more that 3 million lakes, 3000 rivers and 34,000 miles of coastline.  It is one of the most bountiful fishing regions in the world.
However you travel in Alaska you will find great Shopping: Wherever you go you’ll find unique Alaskan products and crafts.  You will want to look for gold nugget jewelry, items carved from jade or handmade clothing and toys.  And don’t forget those famous Alaskan delicacies like canned and smoked salmon, wild berry products and reindeer sausage.
So as your thinking about the best way to see Alaska consider that the best way to see the coastline is by ferry or cruise ship but there are certain types of wildlife you may only see by taking the open road.   And if you want a bird’s eye view of the glaciers maybe a flightsee or day cruise is your thing.
